目的:观察中药复方健脾疏肝方对CCl4诱导小鼠肝纤维化的抑制作用,并探寻其作用机制。方法:将80只雄性清洁级C57BL/6J小鼠随机均分为4组,即正常组、模型组、健脾疏肝组及活血化瘀组。除正常组外,其余3组腹腔注射20%CCl4橄榄油剂1 mL/kg(2次/周,共8周)。正常组腹腔注射等量100%橄榄油剂(2次/周,共8周)。第5周起健脾疏肝组及活血化瘀组分别给予健脾疏肝方和扶正化瘀胶囊灌胃(1次/d,共4周)。实验结束后检测小鼠肝组织基质金属蛋白酶(MMP-1)及其抑制物(TIMP-1)、血清透明质酸(HA)、层黏连蛋白(LN)、三型前胶原(PCⅢ)及四型胶原(Ⅳ-C)水平,采用光镜观察小鼠肝脏病理组织学改变。结果:健脾疏肝组、活血化瘀组TIMP-1、HA、LN、PCⅢ、Ⅳ-C水平均显著低于模型组(P<0.01);健脾疏肝组TIMP-1、HA、LN、PCⅢ、Ⅳ-C水平均显著低于活血化瘀组(P<0.01);与空白组相比,其余3组MMP-1均明显升高(P<0.01),但彼此无明显差异(P>0.05)。结论:健脾疏肝方对CCl4诱导的小鼠肝纤维化具有良好的治疗作用。
Objective: To observe the inhibitory effect of Jianpi Shugan Recipe on CCl4-induced hepatic fibrosis in mice and explore its mechanism. Methods: Eighty male C57BL / 6J mice were randomly divided into 4 groups: normal group, model group, Jianpi Shugan group and Huoxuehuayu group. In addition to the normal group, the remaining three groups were injected intraperitoneally with 20% CCl4 olive oil 1 mL / kg (2 times / week for 8 weeks). The normal group was injected intraperitoneally with the same amount of 100% olive oil (2 times / week for 8 weeks). From the fifth week, Jianpi Shugan group and Huoxuehuayu group were given Jianpi Shugan decoction and Fuzhenghuayu capsule respectively (1 time / d for 4 weeks). After the experiment, the levels of matrix metalloproteinase (MMP-1) and its inhibitor (TIMP-1), serum hyaluronic acid (HA), laminin (LN), type III procollagen Type Ⅳ collagen (Ⅳ-C) levels, the use of light microscopy to observe the pathological changes in liver tissue of mice. Results: The levels of TIMP-1, HA, LN, PCⅢ and Ⅳ-C in Jianpi Shugan group and Huoxuehuayu group were significantly lower than those in model group (P <0.01) (P <0.01). Compared with the blank group, the other three groups of MMP-1 levels were significantly increased (P <0.01), but there was no significant difference (P > 0.05). Conclusion: Jianpi Shugan Decoction has a good therapeutic effect on CCl4-induced hepatic fibrosis in mice.
