为观察IL 12对不同临床分型慢性乙型肝炎患者外周血单个核细胞 (PBMC )产生Th1/Th2类细胞因子的影响。本实验分离 72例慢性乙型肝炎患者 (轻、中、重度 )PBMC ,分别与植物血凝素 (PHA ) (10 0 μg/ml)、HBcAg (1μg/ml)、HBeAg (1μg/ml)单独或联合IL 12 (10ng/ml)体外培养 4 8h ,ELISA法检测培养上清液中IFN γ、IL 10水平。 2 0例健康人群做对照。结果发现IL 12对PHA、HBcAg/HBeAg诱导健康人群PBMC产生Th1/Th2类细胞因子无协同效应 ,对慢性乙型肝炎患者PBMC产生IFN γ有显著协同增殖效应 ,慢性中度患者最为突出。同HBeAg联合诱导 ,不但显著增强慢性乙型肝炎患者PBMC产生IFN γ ,还抑制IL 10的产生。提示IL 12可增强慢性乙型肝炎患者IFN γ水平增高 ,但不同临床分型患者其增殖效应的强度不同 ;IL 12可促进HBeAg诱导的Th2型优势表达向Th1型优势表达转换
To investigate the effects of IL-12 on Th1 / Th2 cytokines production by peripheral blood mononuclear cells (PBMCs) from patients with different clinical classification of chronic hepatitis B patients. In this study, PBMC from 72 patients with chronic hepatitis B (mild, moderate and severe) were isolated and compared with those of PHA (100 μg / ml), HBcAg (1 μg / ml) and HBeAg Or combined with IL 12 (10ng / ml) for 48 h in vitro. The levels of IFN-γ and IL-10 in the culture supernatant were detected by ELISA. 20 healthy controls. The results showed that IL 12 had no synergistic effect on the production of Th1 / Th2 cytokines by PBMCs from PHA and HBcAg / HBeAg-induced PBMCs and IFN-γ production from PBMC from chronic hepatitis B patients. Induction with HBeAg not only significantly enhanced the production of IFN-γ by PBMC in patients with chronic hepatitis B, but also inhibited the production of IL-10. The results suggest that IL 12 may enhance the IFN γ level in patients with chronic hepatitis B, but different clinical types of patients with different intensity of their proliferative effects; IL 12 can promote HBeAg-induced predominant Th2 expression to Th1 predominant expression conversion
