目的 探讨重组人粒细胞集落刺激因子 (rhuG -CSF)对K5 62细胞凋亡和其细胞毒药物敏感性的影响。方法 采用流式细胞术和MTT法分别测定细胞凋亡和细胞毒药物敏感性。结果 在低血清培养条件下 ,rhuG -CSF对K5 62细胞的凋亡有抑制作用 ,其凋亡百分率与rhuG -CSF浓度呈负线性相关 ;rhuG -CSF可增加K5 62细胞对Ara -C的敏感性 ,而降低其对DNR的敏感性 ;在Ara -C和DNR联合条件下 ,rhuG -CSF能增强两药的联合细胞毒毒性 ,克服了单用DNR条件下 ,其使DNR对K5 62细胞毒毒性作用降低的情况。结论 虽然rhuG -CSF并不降低DNR和Ara -C对K5 62细胞的联合细胞毒毒性 ,但当其和化疗药物同时使用治疗髓系白血病患者时 ,仍应慎重。
Objective To investigate the effect of recombinant human granulocyte colony-stimulating factor (rhuG-CSF) on apoptosis and cytotoxicity of K562 cells. Methods Flow cytometry and MTT were used to determine apoptosis and cytotoxicity respectively. Results Under low serum culture conditions, rhuG-CSF inhibited the apoptosis of K562 cells and the percentage of apoptosis was negatively correlated with rhuG-CSF concentration. RhuG-CSF increased the sensitivity of K562 cells to Ara-C And decreased its sensitivity to DNR. Under the combination of Ara-C and DNR, rhuG-CSF could enhance the combined cytotoxicity of the two drugs and overcome the inhibitory effect of DNR on K562 cytotoxicity Reduced toxic effects. Conclusions Although rhuG-CSF does not reduce the combined cytotoxicity of DNR and Ara-C to K562 cells, it should be used with caution when used concurrently with chemotherapeutic agents in patients with myeloid leukemia.
