目的观察蛋白激酶C-ζ(PKC-ζ)通过上皮-间质转化(EMT)对肺腺癌侵袭转移的影响作用。方法应用RNAi技术将合成的小RNA干扰质粒瞬时转染入肺腺癌细胞系A549,进行Western blot检测转染,其后应用Transwell侵袭实验检测细胞体外侵袭能力及转染细胞组中E-cadherin,Vimentin蛋白的表达情况。结果转染72h后,与Scr/A549相比,Si PKC-ζ/A549中PKC-ζ蛋白表达量明显降低,同时Si PKC-ζ/A549的体外侵袭能力明显减弱(P<0.05),并伴有Vimentin蛋白表达水平的降低和E-cadherin蛋白表达水平的升高(P<0.05)。结论 PKC-ζ通过促进肺腺癌细胞EMT的发生,从而促进肺腺癌细胞的侵袭和转移。
Objective To investigate the effect of protein kinase C-ζ (PKC-ζ) on the invasion and metastasis of lung adenocarcinoma through epithelial-mesenchymal transition (EMT). Methods The siRNA plasmid was transiently transfected into lung adenocarcinoma cell line A549 by RNA interference (RNAi) technology. Western blot was used to detect the transfection efficiency. Transwell invasion assay was used to detect the in vitro invasion ability and the expression of E-cadherin, Vimentin protein expression. Results Compared with Scr / A549, the expression of PKC-ζ protein in Si PKC-ζ / A549 was significantly decreased at 72 h after transfection, and the invasive ability of Si PKC-ζ / A549 in vitro was significantly decreased (P <0.05). There was a decrease of Vimentin protein expression and E-cadherin protein expression (P <0.05). Conclusion PKC-ζ promotes the invasion and metastasis of lung adenocarcinoma cells by promoting the occurrence of EMT in lung adenocarcinoma cells.
