目的:研究含CpG序列的寡核苷酸(unmethylated CpG motif containing oligodeoxynucleotide,CpG ODN)对人肺腺上皮细胞β防御素-2(humanβ-defensin2,hBD-2)表达的影响及其作用途径。方法:培养人肺腺上皮细胞A549,用不同浓度的CpG ODN进行干预。用RT-PCR法和Western Blot法检测刺激后A549细胞hBD-2mRNA和蛋白表达变化及细胞核内NF-κB p65蛋白含量的变化。结果:CpG ODN刺激可以促进A549细胞核内NF-κB p65含量增加,活化NF-κB途径,从而诱导hBD-2的表达,与对照组比较P<0.05;用特异性阻断剂阻断NF-κB活化可以部分抑制CpG ODN的诱导作用。结论:CpG ODN可诱导hBD-2在呼吸道上皮细胞的表达,增加机体对外界微生物的抵抗能力,激活NF-κB信号途径可能是其作用机制之一。
AIM: To investigate the effect of CpG ODN on the expression of human β-defensin 2 (hBD-2) and its pathway in human lung epithelial cells. Methods: Human lung adenocarcinoma A549 cells were cultured and treated with different concentrations of CpG ODN. The mRNA and protein expression of hBD-2 and the content of NF-κB p65 protein in A549 cells were detected by RT-PCR and Western Blot. Results: CpG ODN stimulation could promote the increase of NF-κB p65 and the activation of NF-κB in A549 cells, and induce the expression of hBD-2, compared with the control group (P <0.05); Blockade of NF-κB Activation can partially inhibit the induction of CpG ODN. CONCLUSION: CpG ODN can induce the expression of hBD-2 in the airway epithelial cells and increase the resistance of the body to microorganisms. Activation of NF-κB signaling pathway may be one of its mechanisms.
