目的 :观察三氧化二砷 (AS2 O3)对肺癌细胞株NCI H6 9的增殖抑制作用并初步探讨其机制。方法 :肺癌细胞株NCI H6 9用不同浓度AS2 O3处理。MTT法观察不同浓度AS2 O3作用不同时间后对NCI H6 9生长的影响 ;增殖细胞核抗原 (PCNA)单抗LSAB免疫组化染色 流式细胞仪检测PCNA阳性细胞百分比。结果 :AS2 O3明显抑制肺癌细胞株NCI H6 9细胞增殖 ,其作用随AS2 O3浓度增加和作用时间延长而增强 ;随AS2 O3浓度增加 ,PCNA阳性细胞百分比逐渐下降。结论 :AS2 O3可明显抑制肺癌细胞株NCI H6 9的生长 ,调控DNA合成及细胞增殖的PCNA蛋白合成降低可能是AS2 O3的作用机制之一。
Objective: To observe the inhibitory effect of arsenic trioxide (AS2 O3) on lung cancer cell line NCI H6 9 and to explore its mechanism. Methods: Lung cancer cell line NCI H6 9 was treated with different concentrations of AS2O3. MTT assay was used to observe the effect of different concentrations of AS2O3 on the growth of NCI H6 9; Proliferating cell nuclear antigen (PCNA) monoclonal antibody LSAB immunohistochemical staining The percentage of PCNA-positive cells was detected by flow cytometry. Results: AS2 O3 inhibited the proliferation of lung cancer cell line NCI H6 9 cells. The effect of AS2 O3 increased with the increase of AS2 O3 concentration and the prolonged action time. With the increase of AS2 O3 concentration, the percentage of PCNA positive cells gradually decreased. CONCLUSION: AS2 O3 can significantly inhibit the growth of lung cancer cell line NCI H6 9. Regulating PCNA protein synthesis and regulating cell proliferation may be one of the mechanisms of action of AS2 O3.
