目的探讨大鼠视神经挤压伤后及修复过程中睫状神经营养因子(ciliary neuro-trophic factor,CNTF)在视网膜上表达的变化及复方麝香注射液对其影响。方法 72只健康SD大鼠,随机分为正常对照组、模型对照组、药物治疗组3组,每组24只。模型对照组和药物治疗组大鼠均采用钳夹法建立视神经挤压伤实验模型,药物治疗组大鼠腹腔注射复方麝香注射液,正常对照组和模型对照组大鼠腹腔注射等容量的灭菌注射用水,分别于造模后第5、15、30d随机抽取各组8只大鼠处死,摘取眼球行视网膜中CNTF的免疫组化检测。结果实验证实正常视网膜中存在CNTF的表达;视神经损伤后5d、15d、30dCNTF表达均增高,伤后第5d最高,15d下降,但仍明显高于正常对照组(P<0.05),至30d时其表达量与正常对照组已无显著性差异(P>0.05);药物治疗组大鼠视神经损伤后5d、15d、30dCNTF表达均明显高于模型对照组(P<0.05),至伤后治疗15dCNTF表达显著高于模型对照组(P<0.01)。结论视神经损伤后视网膜中CNTF表达增加,复方麝香注射液能有效地促进视网膜中CNTF较长时间高表达,这可能是其对视网膜神经节细胞轴突损伤后的保护作用的机理之一。
Objective To investigate the changes of ciliary neuro-trophic factor (CNTF) expression on retina and the effect of compound musk injection on the retina after crush injury and repair of optic nerve in rats. Methods 72 healthy SD rats were randomly divided into normal control group, model control group and drug treatment group, with 24 rats in each group. Model control group and drug treatment group rats were used to establish the experimental model of optic nerve crush injury, the drug treatment group rats intraperitoneal injection of compound musk injection, normal control group and model control group, intraperitoneal injection of the same volume of sterilization Water was injected into the rats and 8 rats in each group were sacrificed on the 5th, 15th and 30th days after the model was established. All the rats were sacrificed and the immunohistochemistry of CNTF in the retina was performed. Results The expression of CNTF was found in the normal retina. The expression of CNTFF increased 5 days, 15 days and 30 days after optic nerve injury. The expression of CNTFF increased on the 5th day and decreased on the 15th day after injury, but still significantly higher than that of the normal control group (P <0.05) (P> 0.05). The expression of CNTF on the 5th, 15th and 30th day after the optic nerve injury in the drug-treated group was significantly higher than that in the model control group (P <0.05), and the expression of CNTF in the 15th day after injury was significantly higher than that in the normal control group Significantly higher than the model control group (P <0.01). Conclusion The expression of CNTF in the retina increases after optic nerve injury. Compound musk injection can effectively promote the long term expression of CNTF in the retina, which may be one of the protective mechanisms of retinal ganglion cells after axonal injury.
