研究HCVNS5基因的结构与功能,为进一步研制适合我国使用的第3代抗-HCV试剂打下基础。方法:自行设计引物,从河北固安县慢性丙型肝炎患者血中,用逆转录-聚合酶链反应扩增出HCVNS5基因,并将其克隆到载体pKPL-3α,经Dotblot、Southernblot和限制性内切酶分析筛选出阳性克隆pKHC-5α,进行序列测定。结果:核苷酸序列分析表明,克隆的HCVNS5基因与HCVⅡ/1b的核苷酸同源性约90%。结论:所克隆的HCV属于我国常见的HCVⅡ/1b基因型。
To study the structure and function of HCVNS5 gene and lay the foundation for further development of the 3rd generation anti-HCV reagent suitable for our country. Methods: HCV NS5 gene was amplified by reverse transcription polymerase chain reaction from the blood of chronic hepatitis C patients in Gu’an County, Hebei Province. The HCV NS5 gene was cloned into the vector pKPL-3α. Dotblot, Southern blot and restriction endonuclease Dikinase analysis screened positive clone pKHC-5α, the sequence determination. RESULTS: Nucleotide sequence analysis revealed approximately 90% nucleotide homology between the cloned HCV NS5 gene and HCV II / 1b. Conclusion: The cloned HCV belongs to the HCV Ⅱ / 1b genotype in China.
