目的我们前期研究结果表明灯盏花素脂质体前体(PS)减少大脑中动脉栓塞引起的脑梗死面积,改善脑缺血再灌注损伤作用明显优于灯盏花素片(BreT),本文对PS和BreT对血栓形成和溶栓作用进行比较研究。方法线栓法制作大脑中动脉阻塞脑缺血-再灌注损伤模型,经2,3,5-氯化三苯基四氮唑(TTC)染色,检测大鼠脑梗死面积;建立大鼠动-静脉旁路血栓模型、下腔静脉血栓模型、颈总动脉血栓溶栓模型检测血栓湿重;毛细玻管法检测大鼠血小板数目及小鼠凝血时间。结果 PS及BreT均显著减少脑梗死面积(P<0.05),PS作用强于同等剂量的BreT(P<0.05)。PS及BreT组均不同程度降低大鼠动-静脉旁路、下腔静脉及颈总动脉血栓湿重。血栓形成抑制率及溶栓率,PS作用强于同等剂量的BreT。PS及BreT显著延长小鼠凝血时间(P<0.01),PS与同等剂量的BreT相比具有显著性差异(P<0.01)。各组给药前后血小板数量无显著性差异(P>0.05)。结论灯盏花素脂质体前体抑制血栓形成和溶栓作用明显优于灯盏花素片,该作用与其增强抗脑缺血-再灌注损伤作用有关。
Objective Our previous studies showed that breviscapine liposome precursor (PS) reduced the cerebral infarct size caused by middle cerebral artery occlusion and improved cerebral ischemia-reperfusion injury. The effect was better than Brettonine Tablet (BreT). Thrombolysis and thrombolysis were compared with BreT. METHODS: The middle cerebral artery occlusion model of cerebral ischemia-reperfusion injury was established by thread embolization method. The infarct area of rats was detected by 2, 3, 5-trichlorophenyltetrazolium chloride (TTC) staining. The venous bypass thrombus model, inferior vena cava thrombosis model, common carotid thrombosis thrombolysis model were used to detect the thrombus wet weight; the capillary plate method was used to detect the number of platelets in rats and the clotting time in mice. Results Both PS and BreT significantly decreased the cerebral infarct size (P<0.05). The PS effect was stronger than that of the same dose of BreT (P<0.05). Both PS and BreT reduced the arterial venous bypass, inferior vena cava, and common carotid artery thrombus wet weight in different degrees. Thrombosis inhibition rate and thrombolysis rate, PS effect stronger than the same dose of BreT. PS and BreT significantly prolonged clotting time in mice (P<0.01), and PS had significant difference compared with BreT of the same dose (P<0.01). There was no significant difference in platelet counts before and after administration (P>0.05). Conclusion Breviscapine liposome precursor inhibited thrombosis and thrombolysis were significantly better than breviscapine tablets, and this effect was related to its enhancement of anti-cerebral ischemia-reperfusion injury.
