目的研究低氧(2%氧)对成年Wistar大鼠心脏成纤维细胞表型的影响。方法分离培养成年Wistar大鼠心脏成纤维细胞,采用免疫细胞化学染色检测肌成纤维细胞的典型标记———α平滑肌肌动蛋白,用液体闪烁计数方法检测心脏成纤维细胞胶原酶敏感的3H脯氨酸掺入率来观察细胞胶原蛋白合成变化,并检测3H亮氨酸掺入率观察细胞总蛋白合成的变化。结果低氧24h、48h时,成年Wistar大鼠心脏成纤维细胞的α平滑肌肌动蛋白表达较常氧组显著增强;同时细胞胶原蛋白合成速率亦显著增加,在低氧第24h、48h胶原酶敏感的3H脯氨酸掺入率较常氧组分别增加132%(P<0.05)和163%(P<0.01)。结论低氧能够促进成年Wistar大鼠心脏成纤维细胞向肌成纤维细胞表型转换。
Objective To investigate the effect of hypoxia (2% oxygen) on the phenotype of cardiac fibroblasts in adult Wistar rats. Methods Adult Wistar rat cardiac fibroblasts were isolated and cultured. Immunofluorescence staining was used to detect α-smooth muscle actin, a typical marker of myofibroblasts. The cardiac fibroblast collagenase-sensitive 3H mRNA was detected by liquid scintillation counting Tryptophan to observe the change of cell collagen synthesis, and detect the incorporation rate of 3H-leucine to observe the change of cell total protein synthesis. Results Hypoxia 24h, 48h, adult Wistar rat cardiac fibroblasts α-smooth muscle actin expression was significantly increased compared with normoxic group; at the same time, the rate of cellular collagen synthesis also increased significantly 24h, 48h hypoxia collagenase-sensitive 3H proline incorporation increased by 132% (P <0.05) and 163% (P <0.01), respectively, compared with normoxia group. Conclusion Hypoxia can promote the phenotypic transition from cardiac fibroblasts to myofibroblasts in adult Wistar rats.
