目的 :探讨柴龙解郁丹对焦虑模型大鼠行为学的影响及其作用机制。方法 :采用不确定性空瓶刺激法建立大鼠焦虑模型,给予柴龙解郁丹干预1个月并以西酞普兰做对照,观察柴龙解郁丹对焦虑模型大鼠焦虑行为及海马单胺类神经递质(5-HT、DA、NE)含量的影响,单胺类神经递质采用酶联免疫吸附方法(ELISA)检测。结果:柴龙解郁丹能够显著改善模型大鼠焦虑行为优于西酞普兰组(P<0.01),降低大鼠海马区5-HT、DA、NE的含量(P<0.05),但对5-HT的调节作用不及西酞普兰(P<0.01),对DA、NE含量的影响功同西酞普兰(P>0.05),降低单胺类神经递质未呈现出量效关系。结论 :柴龙解郁丹有显著的抗焦虑作用,可能与降低焦虑模型大鼠海马单胺类神经递质而发挥抗焦虑作用有关。
Objective: To investigate the effect and its mechanism of Chai Long Jie Yu Dan on behavior of anxiety model rats. Methods: The rat model of anxiety was established by the uncertainty bottle stimulation. The rats were given Chailong Jieyu Dan for 1 month and citalopram as control. The anxiety behavior and monoamine neurotransmitter (5-HT, DA, NE) content of monoamine neurotransmitters using enzyme-linked immunosorbent assay (ELISA) detection. Results: Chailong Jieyu Dan can significantly improve the anxiety behavior of model rats is superior to citalopram group (P <0.01), reduce the rat hippocampus 5-HT, DA, NE content (P <0.05), but 5-HT (P <0.01). The effect on the content of DA and NE was the same as that of citalopram (P> 0.05), and the decrease of monoamine neurotransmitter did not show the dose-response relationship. Conclusion: Chaiyu Jieyu Dan has a significant anti-anxiety effect, which may be related to the reduction of monoamine neurotransmitters in the hippocampus of anxiety rats.
