目的探讨激活葡萄糖脑苷脂酶(GBA)对帕金森病患者皮肤成纤维细胞自噬活性的影响。方法取32例帕金森病患者的皮肤标本进行成纤维细胞体外培养,运用含有雷帕霉素及不含雷帕霉素的GBA培养基分别进行体外培养,用蛋白印迹法检测自噬体的经典标记物微管相关蛋白的表达。结果雷帕霉素处理后Beclin 1和LC3-Ⅱ/LC3-I蛋白的表达水平增高(P<0.05),而a-synuclein表达水平则下降(P<0.05)。结论激活GBA可以强化自噬水平,降低a-synuclein蛋白的表达。
Objective To investigate the effect of activated GBA on the autophagic activity of skin fibroblasts in patients with Parkinson’s disease. Methods Fibroblasts from 32 Parkinson ’s disease patients were cultured in vitro. The cells were cultured in vitro with GBA medium containing rapamycin and rapamycin. The classic autophagosomes were detected by Western blotting. Marker microtubule-associated protein expression. Results The mRNA expression of Beclin 1 and LC3-Ⅱ / LC3-I increased (P <0.05) and the expression of a-synuclein decreased after rapamycin treatment (P <0.05). Conclusion Activation of GBA can enhance the level of autophagy and decrease the expression of a-synuclein protein.
