目的探索解毒化瘀颗粒是否对急性肝衰竭大鼠肝细胞Cadd45a表达具有调控作用。方法以LPS联合D氨基半乳糖构建急性肝衰竭大鼠模型,解毒化瘀颗粒干预治疗,成模后24h处死各组大鼠,分离肝细胞,Western Bolt和实时荧光定量PCR检测肝细胞Gadd45a蛋白和mRNA表达。结果急性肝衰竭大鼠肝细胞Gadd45a蛋白和mRNA表达明显增高(与空白组比较,P<0.05),而解毒化瘀颗粒可下调Gadd45a蛋白和mRNA表达(与模型对照组比较,P<0.05)。结论高表达的Gadd45a参与急性肝衰竭病理进程,解毒化瘀颗粒可下调Gadd45a表达,影响Gadd45a下游生物学效应,可能包括抑制肝细胞凋亡。
Objective To explore whether Jiedu Huayu granules can regulate the expression of Cadd45a in hepatocytes of rats with acute liver failure. Methods LPS and D-galactosamine were used to establish the acute hepatic failure rat model and the treatment of Jiedu Huayu granule was taken. The rats were sacrificed 24 hours after the model was established. The hepatocytes were isolated and the expression of Gadd45a protein and protein were detected by Western-blot and real- mRNA expression. Results The expression of Gadd45a protein and mRNA in liver cells of rats with acute hepatic failure were significantly increased (P <0.05 compared with the blank group), while the expression of Gadd45a protein and mRNA was down-regulated by Jiedu Huayu granules (P <0.05 compared with the model control group). Conclusion High expression of Gadd45a is involved in the pathological process of acute liver failure. Detoxification and Huayu granules can down-regulate the expression of Gadd45a and affect the downstream biological effects of Gadd45a, which may include inhibition of hepatocyte apoptosis.
