建立应用一对单克隆抗体(MoAb)的免疫层析一步法测定hCG。测定在硝酸纤维素膜上进行,上面包被一条抗αhCGMoAb。用胶体金标记抗βhCGMoAb,作为显示试剂。浸入尿样后,试纸条由于形成AbhCGAb胶体金而出现紫红色线。本法灵敏度为25mIU/ml。假阳性率为0;假阴性率为0。干扰物质在高浓度条件下对测定均无明显影响。当促黄体生成素浓度高至500mIU/ml时不出现交叉反应。说明本方法测试准确,性能稳定
To establish a one-step immunochromatographic assay for detecting hCG using a pair of monoclonal antibodies (MoAb). The assay was performed on a nitrocellulose membrane coated with an anti-ahCGMoAb. Anti-βhCGMoAb was labeled with colloidal gold as a display reagent. Immersed in urine, the test strip due to the formation of Ab hCG Ag colloidal gold appeared purple line. The sensitivity of this method is 25mIU / ml. The false positive rate was 0; the false negative rate was 0. Interfering substances in the high concentration of the conditions had no significant effect on the determination. No cross-reaction occurred when luteinizing hormone concentrations were as high as 500 mIU / ml. This method shows that the test is accurate, stable performance
