目的:骨髓是机体重要的造血器官,其中的单个核细胞主要是单核细胞和巨噬细胞,骨髓组织在骨损伤修复过程中也发挥重要的作用.为提高骨形成蛋白(BMP)修复骨缺损的能力,方法:本实验观察了基因重组人骨形成蛋白-2(re-combinanthumanBoneMorphogeneticProtein-2,rhBMP2)对培养骨髓细胞生物学活性的影响.结果:低浓度的rhBMP2(<0.16mg/L)对培养骨髓细胞的碱性磷酸酶活性、蛋白质含量均有明显的促进作用,且呈剂量依赖性,高浓度的rhBMP2(>0.16mg/L)对培养骨髓细胞的增殖也有一定促进作用结论:本研究的结果进一步证实,骨髓细胞也是BMP重要的靶细胞,BMP不仅促进骨髓细胞向成骨细胞的表型转化,而且可以促进骨髓细胞的增殖,因此在使用复合BMP的植骨材料进行骨缺损修复时,如能辅以骨髓细胞植入,将会大大提高骨缺损的修复效果.
OBJECTIVE: Bone marrow is an important hematopoietic organ in the body. Mononuclear cells are mainly monocytes and macrophages, and bone marrow tissue plays an important role in the process of bone injury repair. In order to improve the ability of bone morphogenetic protein (BMP) to repair bone defects, we investigated the effect of recombinant human bone morphogenetic protein-2 (rhBMP2) on the biological activity of cultured bone marrow cells. Results: Low concentration of rhBMP2 (<0.16mg / L) significantly promoted alkaline phosphatase activity and protein content of cultured bone marrow cells in a dose-dependent and high concentration rhBMP2 (> 0.16mg / L) also promoted the proliferation of cultured bone marrow cells. Conclusion: The results of this study further confirmed that bone marrow cells are also important target cells of BMP. BMP not only promotes the phenotypic transformation of bone marrow cells into osteoblasts, but also promotes the expression of Proliferation, so the use of composite bone graft material for bone defect repair, if combined with bone marrow cell implantation, will greatly improve the repair of bone defects.
