目的:研究硒酸酯多糖(KSC)对CD_3AK细胞增殖活性和杀瘤活性的增强效应。方法:应用CD_3单抗(CD_3McAb)和小剂量rIL-2从正常外周血单个核细胞中诱生CD_3AK细胞;观察KSC在诱生和扩增阶段对CD_3AK细胞增殖特性(增殖指数、激活率、克隆率和分裂指数)、IL-2R表达水平和对Raji、L_(1210)细胞杀伤活性的影响。结果:KSC(5μg/ml)单独对CD_3AK细胞的增殖无明显作用,但增强IL-2和CD_3McAb的作用效应,CD_3AK细胞的增殖指数、激活率、克隆率和分裂细胞指数分别提高39.3%、12.3%、63.3%和49.5%;IL-2R表达水平增高93.5%;杀瘤活性约提高2倍。KSC>10μg/ml时反呈抑制效应。结论:硒酸酯多糖显著增强CF_3AK细胞的增殖活性和杀瘤活性。
Objective: To study the enhancing effect of selenate polysaccharide (KSC) on the proliferation and tumorigenicity of CD_3AK cells. Methods: CD_3AK cells were induced from normal peripheral blood mononuclear cells by CD_3 McAb (CD_3McAb) and low dose of rIL-2. The proliferation of CD_3AK cells (proliferation index, activation rate, cloning Rate and division index), the level of IL-2R expression and the cytotoxicity on Raji and L 1210 cells. Results: KSC (5μg / ml) alone had no effect on the proliferation of CD_3AK cells, but enhanced the effect of IL-2 and CD_3McAb. The proliferation index, activation rate, clonality rate and dividing cell index of CD_3AK cells increased by 39.3% , 12.3%, 63.3% and 49.5%, respectively. The level of IL-2R was increased by 93.5% and the cytotoxic activity was increased about 2-fold. KSC> 10μg / ml showed anti-inhibitory effect. CONCLUSION: Selenate polysaccharide significantly enhanced the proliferation and tumorigenicity of CF_3AK cells.
