目的:研究Ad5F35-IκBαΔN重组腺病毒对K562/DNR细胞耐药性的逆转作用及相关机制。方法:采用流式细胞仪分析细胞凋亡;NF-κB p65活性分析试剂盒检测K562/DNR细胞NF-κB活性。结果:感染Ad5F35-IκBα△N腺病毒后,K562/DNR细胞NF-κB活性受到明显抑制,从0.224±0.021活性单位/μg核蛋白降至0.132±0.015;凋亡率明显增高,从(10.4±1.5)%升至(43.2±4.8)%。结论:感染Ad5F35-IκBα△N腺病毒后,可通过抑制NF-κB的活性,增加K562/DNR细胞凋亡率,从而逆转K562/DNR细胞的耐药性。
Objective: To investigate the reversal effect of Ad5F35-IκBαΔN recombinant adenovirus on K562 / DNR cell resistance and its related mechanisms. Methods: Apoptosis was detected by flow cytometry. The activity of NF-κB in K562 / DNR cells was detected by NF-κB p65 activity assay kit. Results: After infected with Ad5F35-IκBαΔN adenovirus, the activity of NF-κB in K562 / DNR cells was significantly inhibited from 0.224 ± 0.021 cells / μg of nucleoprotein to 0.132 ± 0.015. The apoptotic rate was significantly increased from (10.4 ± 1.5)% to (43.2 ± 4.8)%. Conclusion: Ad5F35-IκBα △ N adenovirus can reverse the K562 / DNR cell resistance by inhibiting the activity of NF-κB, increasing the apoptosis rate of K562 / DNR cells.
