目的:应用肾复康Ⅱ号联合MSCs移植的方法,研究其对肾间质纤维化大鼠肾功能、HGF及其受体C-Met的作用,探讨肾复康Ⅱ号对MSCs旁分泌功能的作用机制。方法:本研究采用全骨髓贴壁筛选法体外分离、扩增培养大鼠MSCs。30只雄性SD大鼠随机分为三组:模型组、MSCs移植组、肾复康Ⅱ号联合MSCs移植组,肾复康Ⅱ号联合MSCs移植组给予肾复康Ⅱ号405mg/kg·d灌胃,模型组、MSCs移植组灌服等体积生理盐水,连续14天,1次/日。大鼠肾间质纤维化模型应用单侧输尿管结扎术制备。MSCs通过尾静脉注射的方法进行移植。移植后1周、2周、4周,对各组大鼠的肾功能、肾组织中HGF和C-Met进行检测。结果:MSCs移植组、肾复康Ⅱ号联合MSCs移植组与模型组相比,肾功能改善,HGF、C-Met分泌增加;肾复康Ⅱ号联合MSCs移植组肾功能改善更明显,HGF、C-Met分泌明显增加,各组之间比较,具有显著性差异。结论:肾复康Ⅱ号可能是作用于HGF及其受体C-Met调控MSCs旁分泌功能达到改善肾功能的作用。
OBJECTIVE: To investigate the effect of Renfukang II combined with MSCs transplantation on renal function, HGF and its receptor C-Met in renal interstitial fibrosis rats, and to investigate the paracrine function of Renfukang II on MSCs. Mechanism. METHODS: In this study, rat bone marrow MSCs were isolated and expanded in vitro using a whole bone marrow adherence screening method. Thirty male Sprague-Dawley rats were randomly divided into three groups: model group, MSCs transplantation group, Renfukang II combined MSCs transplantation group, and Renfukang II combined MSCs transplantation group given Renfukang II 405mg/kg·d irrigation. The stomach, model group, and MSCs transplantation group were given an equal volume of normal saline for 14 consecutive days, once daily. The rat renal interstitial fibrosis model was prepared by unilateral ureteral ligation. MSCs were transplanted by tail vein injection. At 1 week, 2 weeks, and 4 weeks after transplantation, renal function and renal tissue HGF and C-Met were measured in each group. RESULTS: Compared with the model group, the MSCs transplantation group and Renfukang II combined MSCs transplantation group had improved renal function, increased secretion of HGF and C-Met, and Renfukang II combined with MSCs transplantation group had significantly improved renal function. The secretion of C-Met increased significantly and there was a significant difference between the groups. Conclusion: Shenfukang II may play a role in HGF and its receptor C-Met regulating paracrine function of MSCs to improve renal function.
