目的通过检测不同口腔念珠菌的细胞外代谢产物,绘制不同念珠菌的特征性代谢谱图,探讨基于核磁共振的代谢组学方法在快速鉴别不同念珠菌的种类和分析相关代谢产物的化学物质种类的可能性。方法分别在沙保培养基中接种白色念珠菌、克柔念珠菌、光滑念珠菌、热带念珠菌,采取比浊法计数,绘制生长曲线,取生长活跃期的四种念珠菌培养液,行核磁共振检测和主成分分析,比对webspectra数据库中的化学物质种类的标准谱图,得出相关代谢组分。结果四种念珠菌的细胞外代谢产物具有各自的类聚关系,主成分分析法可以区分这四种念珠菌;四种念珠菌的代谢组化学物质种类不同,可能与不同念珠菌菌种有关。结论基于核磁共振的代谢组学检测方法在口腔念珠菌的快速检测中和相关代谢产物的分析中有很好的应用前景。
OBJECTIVE: To detect the characteristic metabolic profiles of Candida different Candida species through the detection of different Candida albicans extracellular metabolites, to explore the method based on nuclear magnetic resonance metabonomics in the rapid identification of different Candida species and analysis of metabolites related to chemical species The possibility of. Methods Candida albicans, Candida krusei, Candida glabrata and Candida tropicalis were inoculated into Sabouraud culture medium respectively. The turbidimetric method was used to count the growth curves. Four kinds of Candida culture broths with active growth stage were obtained. Resonance detection and principal component analysis were used to compare the standard spectra of chemical species in the webspectra database to derive the relevant metabolic components. Results The extracellular metabolites of Candida albicans had their own clustering relationship. The principal component analysis could distinguish the four Candida species. The metabolites of Candida albicans differed in the types of chemical substances and may be related to different Candida species. Conclusion The method of metabonomics based on nuclear magnetic resonance has a good application prospect in the rapid detection of oral Candida and the analysis of related metabolites.
