目的:观察鬼针草总黄酮对大鼠内毒素性急性肺损伤(ALI)的抗炎和抗氧化作用,探讨其对ALI保护作用的机制。方法:120只大鼠随机分为正常对照组(生理盐水),模型组,地塞米松组(5 mg.kg-1,ip),鬼针草总黄酮低、中、高剂量组(100,150,200 mg.kg-1,ip),每组20只。给药3 d,末次ip给药1 h后,ip内毒素诱导ALI模型,6 h后处死,观察每组肺泡灌洗液白细胞和蛋白含量、肺组织核转录因子(NF-κB)p65表达以及肿瘤坏死因子(TNF-α)、白细胞介素(IL-6)、白细胞介素(IL-10)、超氧化物歧化酶(SOD)、丙二醛(MDA)含量以及中性粒细胞髓过氧化物酶(MPO)活性。结果:与对照组比较,模型组的肺泡灌洗液白细胞、蛋白含量显著增加(P<0.05~P<0.01);鬼针草总黄酮中、高剂量组肺组织NF-κB p65表达为(38.46±4.56)%,(31.39±4.18)%,MDA含量为(1.73±0.19),(1.46±0.15)nmol.mg-1,TNF-α含量为(259.33±37.48),(211.46±32.69)μg.L-1,IL-6含量为(287.46±100.18),(223.55±93.49)ng.L-1,MPO活性为(2.69±0.57),(2.43±0.38)U.g-1,与模型组比较均明显降低(P<0.05~P<0.01);IL-10含量为(16.73±3.87),(17.28±3.58)μg.L-1,SOD为(69.46±9.84,73.24±8.39)U.mg-1,与模型组比较均明显升高(P<0.05~P<0.01)。结论:鬼针草总黄酮能减轻LPS诱发急性肺损伤所致肺血管通透性,减少炎症渗出,降低氧化应激损伤程度。
Objective: To observe the anti-inflammatory and anti-oxidative effects of the total flavonoids of Bidens bipinnata L. on acute lung injury induced by endotoxin in rats, and to explore its mechanism of protective effect on ALI. Methods: 120 rats were randomly divided into normal control group (saline), model group, dexamethasone group (5 mg.kg-1, ip), low, medium and high dose kg-1, ip), 20 in each group. After 3 d of the last ip administration, the model of ALI was induced by ip endotoxin and then killed after 6 h. The contents of leukocyte and protein in lung lavage fluid and expression of nuclear factor-kappa B (NF-κB) p65 in each group were observed The levels of TNF-α, IL-6, IL-10, SOD, MDA and the myelocystis of neutrophils Oxidase (MPO) activity. Results: Compared with the control group, leukocyte and protein contents in the alveolar lavage fluid of the model group were significantly increased (P <0.05 ~ P <0.01). The expression of NF-κB p65 in the lung tissue of middle and high dosage of Bidens pilosa was (38.46 (31.39 ± 4.18)%, MDA content was (1.73 ± 0.19) and (1.46 ± 0.15) nmol.mg-1 respectively. The content of TNF-α was (259.33 ± 37.48) and (211.46 ± 32.69) μg respectively. The levels of IL-6 and IL-6 in serum were (287.46 ± 100.18) and (223.55 ± 93.49) ng.L-1, respectively. MPO activity was (2.69 ± 0.57) and (2.43 ± 0.38) (P <0.05 ~ P <0.01). The content of IL-10 was (16.73 ± 3.87), (17.28 ± 3.58) μg.L-1 and the SOD was (69.46 ± 9.84,73.24 ± 8.39) Compared with the model group were significantly increased (P <0.05 ~ P <0.01). CONCLUSION: The total flavonoids of Bidens bipinnata can reduce pulmonary vascular permeability induced by LPS-induced acute lung injury, reduce inflammatory exudation and reduce the degree of oxidative stress injury.
