目的:通过对传统精子染色质扩散实验(SCD)方法进行改良,探索一种更简单、快速、准确的精子DNA完整性检测方法。同时对改良SCD法检测的正常参考值范围进行初步探讨。方法:对传统SCD法在实验过程、试剂配制及器具使用上进行了改良优化,同时对改良前后SCD法进行了比较。此外,利用改良SCD法对293例正常生育男性精液进行精子DNA完整性检测。结果:建立了改良SCD检测法,经统计分析改良前后测定结果无统计学差异,同时初步对改良SCD法检测的正常精子DNA百分比参考值范围进行了确定,其值为精子DNA异常发生率<32.58%。结论:改良SCD方法能更快速地对精子DNA的完整性进行检测,同时,由于降低了试剂成本,该法更容易在实际工作中得到普及。
OBJECTIVE: To explore a more simple, rapid and accurate method for the detection of DNA integrity in sperm by improving the traditional sperm chromatin diffusion test (SCD). At the same time improve the SCD method to detect the normal reference range for a preliminary discussion. Methods: The traditional SCD method was modified and optimized in the process of experiment, preparation of reagents and the use of apparatus. At the same time, the SCD method was compared before and after the improvement. In addition, the sperm DNA integrity of 293 normal fertile male sperm was tested by modified SCD method. Results: The improved SCD method was established. There was no statistical difference between the two methods before and after the improvement by statistical analysis. Meanwhile, the reference value range of normal sperm DNA was determined by modified SCD method. The value of sperm DNA abnormality was <32.58 %. Conclusion: The improved SCD method can detect the DNA integrity of sperm more quickly, and at the same time, it is easier to be popularized in practice due to lower cost of reagents.
