目的:实验观察阴道毛滴虫十堰分离株对灭滴灵的敏感性。方法:收集阴道毛滴虫临床分离株,体外传代培养4代,培养11管至对数生长期,调整虫体浓度为0.5×106个/ml,计算加入甲硝唑,使终浓度分别为:62.50μg/ml、50.00μg/ml、31.25μg/ml、25.00μg/ml、15.63μg/ml、7.81μg/ml、3.91μg/ml、1.95μg/ml、0.98μg/ml、0.49μg/ml、0μg/ml,置37℃培养,分别于2、4、6、8、12、24 h观察虫体生长存活情况,计算高倍镜下虫体数目和死亡率。结果:62.50μg/ml和50.00μg/ml浓度组,滴虫死亡率均为100.00%;作用后2 h,15.63μg/ml浓度组死亡率为7.69%,31.25μg/ml组死亡率为65.51%;作用后4 h,7.81μg/ml浓度组死亡率为78.94%,3.91μg/ml浓度组死亡率为18.75%;作用后6 h,3.91μg/ml浓度组死亡率为73.68%,1.95μg/ml浓度组死亡率为21.52%;24 h滴虫最低致死量为3.91μg/ml。结论:阴道毛滴虫十堰分离株对灭滴灵较敏感,体外有效最低杀虫浓度为3.91μg/ml。
Objective: To observe the sensitivity of Trichomonas vaginalis isolates to metronidazole. Methods: The clinical isolates of Trichomonas vaginalis were collected and subcultured for 4 passages in vitro. Eleven tubes were cultured to logarithmic growth phase. The concentration of parasites was adjusted to 0.5 × 106 / ml. Metronidazole was added to calculate the final concentration of Trichomonas vaginalis. 50 μg / ml, 31.25 μg / ml, 25.00 μg / ml, 15.63 μg / ml, 7.81 μg / ml, 3.91 μg / ml, 1.95 μg / ml, 0.98 μg / ml, 0.49 μg / 0μg / ml, set at 37 ℃ culture, respectively, 2,4,6,8,12,24 h observe the growth and survival of parasites, calculate the number of high magnification worms and mortality. Results: The mortality of trichomoniasis was 100.00% in the concentration of 62.50μg / ml and 50.00μg / ml respectively. The mortality of 15.63μg / ml concentration group was 7.69% and the mortality rate of 31.25μg / ml group was 65.51% ; At 4 h after the treatment, the mortality rate was 78.94% in 7.81μg / ml group and 18.75% in 3.91μg / ml group; the mortality rate in 3.91μg / ml group was 73.68% and 1.95μg / ml concentration of 21.52% mortality rate; 24 h Trichomonas minimum lethal dose of 3.91μg / ml. Conclusion: Trichomonas vaginalis isolates are sensitive to metronidazole, and the lowest effective insecticidal concentration in vitro is 3.91μg / ml.
