目的 :运用基因表达谱芯片和原位杂交技术观察人 PTN(pleiotrophin)基因在某些恶性肿瘤中的表达 ,并探讨其在肿瘤发生发展中的作用。 方法 :用 Bio Door 40 96型 c DNA基因表达谱芯片和原位杂交技术检测 PTN基因在 5例脑胶质瘤、10例喉鳞癌、6例肝细胞癌及正常对照组织中的表达水平。 结果 :在上述肿瘤的基因表达谱芯片检测中 ,PTN基因表达均显著增高 ,与脑胶质瘤、喉鳞癌、肝细胞癌的原位杂交检测结果相吻合。 结论 :c DNA基因表达谱芯片能够为恶性肿瘤的相关基因功能分析提供特异和可靠的数据。 PTN基因在恶性肿瘤的发生机制中可能起重要作用。
Objective: To use gene expression microarray and in situ hybridization to observe the expression of human PTN (pleiotrophin) gene in some malignant tumors, and to explore its role in tumor development. Methods : The expression level of PTN gene in 5 cases of glioma, 10 cases of laryngeal squamous cell carcinoma, 6 cases of hepatocellular carcinoma and normal control tissues were detected with Bio Door 40 96 c DNA gene expression microarray and in situ hybridization. RESULTS : In the above gene expression microarray analysis of tumors, PTN gene expression was significantly increased, consistent with the in situ hybridization results of glioma, laryngeal squamous cell carcinoma and hepatocellular carcinoma. Conclusion : c DNA gene expression microarray can provide specific and reliable data for the analysis of related genes in malignant tumors. PTN gene may play an important role in the pathogenesis of malignant tumors.
