移植物抗宿主病(GVHD)是骨髓移植中的主要难题之一。动物实验表明,除去骨髓移植物中的成熟T淋巴细胞,可消除这种急性GVH病。本文介绍一种新的大鼠单克隆抗体(McAb),能与人补体结合,可以消除人移植骨髓中的成熟T淋巴细胞。 CAMPATH1McAb是用富含T淋巴细胞的正常人外周血单个核细胞免疫ADA大鼠,然后取脾细胞与Y3Ag1,2,3大鼠骨髓瘤细胞系融合获得的。用酶联、间接免疫荧光和微量细胞毒试验进行筛选,发现CAMPATH1有较强的细胞毒作用,属于IgM。作者用人补体做细胞毒试验结合免疫荧光及细胞荧光流图进行了细致的分析,结果证明扁桃腺和胸腺组织中的绝大多数细胞都与CAMPATH1反应,显示CAMP-
Graft-versus-host disease (GVHD) is one of the major challenges in bone marrow transplantation. Animal experiments show that the removal of mature T lymphocytes in bone marrow transplantation can eliminate this acute GVH disease. This article describes a new rat monoclonal antibody (McAb) that binds to human complement and eliminates mature T lymphocytes in human bone marrow. CAMPATH1McAb is an ADA rat immunized with normal human peripheral blood mononuclear cells enriched in T lymphocytes and then fused with Y3Ag1, 2, 3 rat myeloma cell line. Enzyme-linked, indirect immunofluorescence and micro-cytotoxicity test screening, found that CAMPATH1 has a strong cytotoxicity, belonging to IgM.
