目的 研究凋亡基因ras突变和rasP2 1蛋白表达及DNA倍体异常与尿路上皮肿瘤预后关系。方法 应用聚合酶链式反应加单链多态性分析 (PCR SSCP)及流式细胞术和细胞免疫荧光技术检测 52例尿路上皮肿瘤K ras基因突变及rasP2 1蛋白表达和DNA含量变化。K ras基因突变5例 (9 6 % ) ,rasP2 1蛋白阳性表达 36例 (6 9 2 % ) ;异倍体 2 2例 (4 2 3% ) ,二倍体 30例 (57 7% ) ;DNA倍体异常和rasP2 1蛋白表达随肿瘤病理分级和分期的上升而增加 ,与肿瘤预后呈正相关 (P <0 0 1) ;异倍体肿瘤其rasP2 1蛋白表达率明显高于二倍体肿瘤 (P <0 0 1)。结论 DNA倍体异常和rasP2 1蛋白表达对尿路上皮肿瘤预后有预测意义
Objective To study the relationship between ras mutation of apoptotic gene and rasP2 1 protein expression, DNA ploidy abnormality and prognosis of urothelial tumors. Methods Polymerase chain reaction plus single-stranded polymorphism analysis (PCR-SSCP), flow cytometry and cellular immunofluorescence were used to detect the K ras gene mutation, rasP2 1 protein expression and DNA content in 52 cases of urothelial neoplasms. K ras gene mutation was found in 5 cases (96%), rasP21 protein was positively expressed in 36 cases (6.92%), aneuploidy was found in 22 cases (4.23%), diploid in 30 cases (57%). The abnormality of DNA ploidy and rasP2 1 protein increased with the increase of tumor histological grade and staging, which was positively correlated with the prognosis of the tumor (P <0 01). The expression of rasP2 1 protein in aneuploid tumors was significantly higher than that of diploid tumors. (P < 0 0 1). Conclusions The abnormality of DNA ploidy and rasP2 protein expression have predictive value for the prognosis of urothelial tumors
