目的:建立同时测定软蒺藜中异鼠李素-7-O-α-L-吡喃鼠李糖苷和苜蓿素2种黄酮类成分含量的方法。方法:采用HPLC法,用HIQ sil C18W色谱柱(250 mm×4.6 mm,5μm)分离,以甲醇-水(60∶40)为流动相,流速0.8 mL.min-1,检测波长360 nm,柱温30℃。结果:异鼠李素-7-O-α-L-吡喃鼠李糖苷和苜蓿素2种成分的线性范围分别为3.223~80.58μg.mL-1(r=0.9997)和1.720~43.00μg.mL-1(r=0.9995),方法回收率(n=9)分别为100.5%和98.7%。结论:本方法简便、快捷,结果准确,重复性好,可应用于软蒺藜中异鼠李素-7-O-α-L-吡喃鼠李糖苷和苜蓿素2种黄酮类化学成分含量的同时测定。
OBJECTIVE: To establish a method for the simultaneous determination of isorhamnetin-7-O-α-L-rhamnopyrhamnoside and hirudin in soft fleas. Methods: HPLC method was used to separate HIQ sil C18W column (250 mm×4.6 mm, 5 μm) with methanol-water (60∶40) as mobile phase, flow rate of 0.8 mL.min-1, detection wavelength of 360 nm, column Temperature 30 °C. Results: The linear ranges of isorhamnetin-7-O-α-L-rhamnopyranoside and hirudin were 3.223~80.58μg.mL-1(r=0.9997) and 1.720~43.00μg, respectively. mL-1 (r=0.9995), method recovery (n=9) were 100.5% and 98.7%, respectively. Conclusion: The method is simple, rapid, accurate and reproducible. It can be applied to the content of chemical components of Isorhamnetin-7-O-α-L-rhamnopyrhamnoside and Alizarin in soft fleas Simultaneous determination.
