目的检测大肠癌组织中癌基因垂体腺瘤转化基因(PTTG)和原癌基因(c-Myc)蛋白表达情况并探讨其意义。方法采用组织芯片技术和免疫组织化学SP法检测55例大肠癌、15例正常大肠黏膜中两者的表达情况。结果大肠癌组织中PTTG和c-Myc表达定位于细胞质和细胞核,其阳性表达率分别为74%(41/55)和67%(37/55)与正常大肠黏膜组织13%(2/15)、27%(4/15)。两者的表达在大肠癌患者的不同年龄、性别等差异无统计学意义(P>0.05),但在不同病理分化程度、淋巴结转移和临床分期间表达差异有统计学意义(P<0.05);在大肠癌组织中PTTG蛋白的阳性表达与c-Myc蛋白的阳性表达呈正相关(P<0.05)。结论 PTTG和c-Myc在大肠癌组织中的表达情况,提示PTTG高表达可能与肿瘤发生有关,且与其浸润、发展、转移有关,c-Myc与PTTG相互协调,共同在大肠癌的发生发展中发挥重要作用。
Objective To detect the expression of PTTG and c-Myc in colorectal carcinoma and to explore its significance. Methods Tissue microarray technique and immunohistochemical SP method were used to detect the expression of the two proteins in 55 cases of colorectal carcinoma and 15 cases of normal colorectal mucosa. Results The expression of PTTG and c-Myc in colorectal carcinoma was localized in the cytoplasm and nucleus. The positive expression rates of PTTG and c-Myc were 74% (41/55) and 67% (37/55) respectively in normal colorectal mucosa tissues and 13% (2/15) , 27% (4/15). There was no significant difference in the expression of these two genes between different age and sex in patients with colorectal cancer (P> 0.05), but there was significant difference between different pathological grade, lymph node metastasis and clinical stage (P <0.05). The positive expression of PTTG protein in colorectal carcinoma was positively correlated with the positive expression of c-Myc protein (P <0.05). Conclusions The expressions of PTTG and c-Myc in colorectal carcinoma suggest that the high expression of PTTG may be involved in the tumorigenesis, which may be related to its infiltration, development and metastasis. The c-Myc and PTTG coordinate with each other in the development of colorectal cancer Play an important role.
