近年来DNA技术的进展使寄生虫学家们能应用该技术作寄生虫种、株的鉴定。在血吸虫,已有研究证明曼氏血吸虫rDNA的重复单位长约10kb。用BamHI内切酶降解获得三个有完整长度的重复单位片段,并在pBR322中克隆化,制备探针用于其它血吸虫虫种的研究,结果发现了种特异的限制性内切酶位点及非转录区(NTS)长度的种间变异。本文拟用这三个探针研究片形吸虫属中某些虫种rDNA重复单位的种间和种内变异。这些吸虫为:肝片形吸虫(9个样品),巨片形吸虫(2个),Fasciola sp.(1个)和大拟片形吸虫(1个),新鲜冻存或存放在70%乙醇中,分别
In recent years, advances in DNA technology have enabled parasiteologists to apply the technology as parasite species and strain identification. In Schistosoma japonicum, it has been reported that the repeat unit of Schistosoma mansoni rDNA is about 10 kb in length. Three full-length repeat unit fragments were obtained by BamHI endonuclease digestion and cloned in pBR322 to prepare probes for the study of other schistosomiasis species.
