将轮移因子(TF)加入到1,6-二苯基-1,3,5-己三烯(DPH)探针标记的人血淋巴细胞中,测其在不同时间的荧光偏振度。发现加入TF后,淋巴细胞膜的偏振度在第1小时明显降低,即淋巴细胞膜流动性增强。此外,探讨了pH、TF浓度、时间及氯离子对膜流动性的干扰问题。本研究结果显示:(1)膜流动性测定可作为TF作用于淋巴细胞的早期反应指标。(2)膜流动性的变化可能与TF信息传递有关。
The migration factor (TF) was added to human hemocytes labeled with 1,6-diphenyl-1,3,5-hexatriene (DPH), and their fluorescence polarization degree at different time was measured. After adding TF, the degree of polarization of the lymphocyte membrane was significantly reduced at the first hour, that is, the fluidity of the lymphocyte membrane was enhanced. In addition, the interferences of pH, TF concentration, time and chloride ion on membrane fluidity were discussed. The results of this study show: (1) The membrane fluidity assay can be used as an early response indicator of TF on lymphocytes. (2) The change of membrane fluidity may be related to TF information transmission.
