目的研究藤茶提取物藤茶总黄酮(TF)、二氢杨梅素(DMY)及杨梅素(Myr)的抗凝血及溶栓作用。方法采用毛细玻璃管法观察对小鼠凝血时间的影响;采用下腔静脉结扎大鼠血栓模型,观察受试药物的抗血栓作用。结果与正常对照组比较,高剂量(2 mg.kg-1)的TF、DMY及Myr均能明显延长小鼠凝血时间(P<0.05,P<0.01);低剂量(0.5 mg.kg-1)的TF、DMY及Myr虽有延长小鼠凝血时间的趋势,但差异无统计学意义。下腔静脉结扎血栓模型结果显示,阳性药川芎嗪、Myr及DMY各组的血栓干重均明显低于正常对照组(P<0.01,P<0.05);Myr高剂量组的血栓湿重明显低于正常对照组(P<0.01),TF高、低剂量组的血栓湿重、干重与正常对照组比较,差异均无统计学意义。各实验组对血栓的抑制率大小依次为:Myr高剂量组>川芎嗪组>DMY低剂量组>Myr低剂量组>DMY高剂量组>TF高剂量组>TF低剂量组。结论 Myr和DMY可能是藤茶抗血栓作用的主要有效成分,且Myr的抗血栓作用稍强于DMY。
Objective To study the anticoagulant and thrombolytic effects of rattan tea extract TF, DMY and Myr. Methods The effect of blood coagulation time was observed by capillary glass tube method. The model of thrombosis was established by ligating the inferior vena cava and the antithrombotic effect of the tested drugs was observed. Results Compared with the normal control group, high dose (2 mg.kg-1) TF, DMY and Myr could significantly prolong the clotting time (P <0.05, P <0.01) ) TF, DMY and Myr have the tendency to prolong clotting time in mice, but the difference was not statistically significant. The model of inferior vena cava thrombus ligation showed that the thrombus dry weights of the ligustrazine, Myr and DMY positive groups were significantly lower than those of the normal control group (P <0.01, P <0.05), and the thrombus weight of the Myr high dose group was significantly lower In the normal control group (P <0.01), there was no significant difference in the thrombus wet weight and dry weight between the TF high and low dose groups compared with the normal control group. The inhibitory rates of thrombus in each experimental group were as follows: Myr high dose group> Ligustrazine group> DMY low dose group> Myr low dose group> DMY high dose group> TF high dose group> TF low dose group. Conclusion Myr and DMY may be the main anti-thrombotic components of rattan tea, and Myr’s anti-thrombotic effect is slightly stronger than DMY.
