目的建立一种快速检测非O1、非O139群霍乱弧菌方法,并对浙江省部分地区海水产品中非O1、非O139群霍乱弧菌及其携带毒力基因开展快速检测。方法采用三糖斜面、氧化酶实验、粘丝实验、无盐胨水等简单生化进行初筛,对全部结果阳性的菌株用聚合酶链反应(PCR)方法检测霍乱弧菌外膜蛋白OmpW以及毒力基因ctxA、hlyA和toxR。结果按照方法用三糖斜面初筛后共有504份样本阳性,其中氧化酶、粘丝和无盐胨水全部为阳性的有434份;有388份样本OmpW PCR结果为阳性,其中ctxA结果均为阴性,toxR阳性hlyA阳性的有225株。结论建立的快速检测方法提高了非O1、非O139群霍乱弧菌的检出率,缩短了检测时间,节约了检测成本。浙江省非O1、非O139群霍乱弧菌在海水产品中广泛存在,应对其致病性提高警惕,加强监测。
Objective To establish a rapid method for the detection of Vibrio cholerae other than O1 and non-O139 strains, and to rapidly detect non-O1, non-O139 Vibrio cholerae and their virulence genes in marine products in Zhejiang Province. Methods Trifluralin, oxidase, viscidity test, peptone and other simple biochemical tests were carried out to screen all strains with positive results. Polymerase chain reaction (PCR) was used to detect OmpW and Vibrio cholerae OMPW Genes ctxA, hlyA and toxR. Results According to the method, a total of 504 samples were positive after screening with trisaccharide bevel. Among them, 434 samples were positive for oxidase, mucilage and non-salt peptone, and 388 samples were positive for OmpW PCR. The results of ctxA were Negative, toxR positive hlyA positive 225. Conclusion The rapid detection method established can improve the detection rate of non-O1, non-O139 Vibrio cholerae, shorten the detection time and save the cost of detection. Zhejiang province non-O1, non-O139 Vibrio cholerae widespread in marine products, should be vigilant to enhance its pathogenicity and strengthen monitoring.
