目的研究新疆阿勒泰地区图瓦人Y染色体上DYS287、DYS440位点的遗传多态性,比较多个民族这2个位点的遗传多态信息,初步推测新疆阿勒泰地区图瓦人的起源。方法应用聚合酶链反应扩增DYS287和DYS440位点,2%琼脂糖电泳分析50名新疆阿勒泰地区图瓦人男性个体的基因型。应用生物信息学技术,比对多个民族这2个位点的遗传多态信息,计算不同群体间的遗传距离。结果 DYS287位点共检测到YAP+和YAP-2种基因型。检出YAP+13名,其基因型频率为26%。DYS440位点共检测到DYS440*3和DYS440*42种基因型。DYS440*3和DYS440*4的基因型频率分别为58%和42%。结论图瓦人是一个隔离群,是探索起源的理想人群。新疆阿勒泰地区图瓦人和蒙古族、哈萨克族、维吾尔族、乌孜别克族以及塔吉克族可能具有不同的种族起源。
Objective To study the genetic polymorphisms of DYS287 and DYS440 on the Y chromosome of Tuva in Altay, Xinjiang and to compare the genetic polymorphism information of these two loci in several ethnic groups to infer the origin of Tuva in Altay, Xinjiang. Methods The DYS287 and DYS440 loci were amplified by polymerase chain reaction (PCR) and genotypes of 50 male Tuva natives in Altay, Xinjiang were analyzed by 2% agarose gel electrophoresis. Using bioinformatics technology, we compared the genetic polymorphism information of two loci in different ethnic groups and calculated the genetic distance among different populations. Results YAP + and YAP-2 genotypes were detected in DYS287 locus. YAP + 13 were detected with a genotype frequency of 26%. DYS440 locus detected a total of DYS440 * 3 and DYS440 * 42 genotypes. The genotype frequencies of DYS440 * 3 and DYS440 * 4 were 58% and 42% respectively. Conclusion Tuva is a segregation group and an ideal group to explore origins. The Tuva and Mongolian, Kazak, Uighur, Uzbek and Tajik ethnic groups in the Altay area of Xinjiang may have different racial origins.
