目的:检测原发性舍格伦综合征(primary Sjgren’s syndrome,pSS)患者血细胞中干扰素调节因子(IRF)1、3、7的表达水平,并检测IRF1在pSS患者腮腺组织中的表达。方法:采用实时定量PCR,对37例pSS患者和24例对照组患者血细胞中IRF1、3、7的水平进行检测,比较分析其在两组之间的差异。采用免疫组化和免疫荧光法检测IRF1在pSS患者腮腺组织中的表达。应用SAS6.12软件包对数据进行t检验。结果:实时定量PCR法检测结果发现,pSS组的IRF1mRNA表达量显著高于对照组,两组之间有显著差异(P=0.00001),pSS组的IRF1是对照组的2.17倍,而IRF3和IRF7在两组之间无显著差异(P=0.3813,0.4501)。免疫组化和免疫荧光发现,pSS组患者腮腺组织中的导管上皮细胞、淋巴细胞和上皮岛均为IRF1阳性;而对照组腮腺组织中仅导管上皮细胞为IRF1阳性。结论:IRF1在原发性舍格伦综合征患者中存在异常表达。
Objective: To detect the expression of interferon regulatory factor (IRF) 1,3,7 in blood cells of patients with primary Sjögren’s syndrome (pSS) and to detect the expression of IRF1 in the parotid gland of patients with pSS . Methods: The real-time quantitative PCR was used to detect the levels of IRF1, 3 and 7 in the blood cells of 37 pSS patients and 24 control patients. The difference between the two groups was analyzed. The expression of IRF1 in the parotid gland of pSS patients was detected by immunohistochemistry and immunofluorescence. Data was t-tested using the SAS6.12 software package. Results: The results of real-time quantitative PCR showed that the expression of IRF1 mRNA in pSS group was significantly higher than that in control group (P = 0.00001), IRF1 in pSS group was 2.17 times of that in control group, while IRF3 and IRF7 There was no significant difference between the two groups (P = 0.3813, 0.4501). Immunohistochemistry and immunofluorescence revealed that the ductal epithelial cells, lymphocytes and epithelial islands in the parotid gland of the pSS group were both IRF1-positive and the ductal epithelial cells in the parotid gland of the control group were IRF1-positive. Conclusion: IRF1 is abnormally expressed in patients with primary Sjogren’s syndrome.
