龙江血居吸虫 ( Sangunicola lungensis)和日本血吸虫 ( Schistosoma japonicum)依次为冷血动物和温血动物血吸虫的重要代表种类。分别对日本血吸虫与龙江血居吸虫成虫和尾蚴全基因组进行 RAPD标记研究 ,筛选出 1 4种随机引物对 4种材料扩增得到 1 50条多态片段 ,其中日本血吸虫成虫和尾蚴之间的同源性为0 .60 9;龙江血居吸虫成虫和尾蚴之间的同源性为 0 .61 7,两种血吸虫成虫之间的同源性为 0 .1 4 8,尾蚴之间为 0 .1 4 5。说明了冷血和温血动物血吸虫 ,它们具有相近的遗传背景 ,同时揭示了同一种血吸虫 ,其成虫和尾蚴阶段存在一定的多态性 ,反映出在虫体发育过程中 ,基因组的结构可能发生变化。根据 2 8S r RNA特定基因片段的 RAPD分析 ,筛选出对两种血吸虫不同发育阶段具有特异性的扩增引物 ,进一步说明同一种血吸虫在不同发育阶段 2 8S r RNA基因片段存在的差异。
Sangunicola lungensis and Schistosoma japonicum were the most important representative species of cold-blooded animals and warm-blooded animals. RAPD markers were used to study the whole genome of adult and cercariae of Schistosoma japonicum and Longjiang, and 14 random primers were screened to amplify 14 kinds of materials to obtain 150 polymorphic fragments. Among them, the adult Schistosoma japonicum and cercariae The homology was 0.60 9; the homology between adult and cercariae of Longjiang bloodworm was 0.61 7, the homology between the two adult schistosomes was 0.148, the cercariae were 0 .1 4 5. The cold-blooded and warm-blooded animals, Schistosoma japonicum, have similar genetic background and reveal that the same species of Schistosoma japonicum has some polymorphism in its adult and cercarial stages, indicating that the structure of the genome may change during the development of the worms . According to RAPD analysis of specific gene fragments of 2 8S rRNA, primers specific for different developmental stages of the two schistosomes were screened out to further elucidate the differences of the 2 8S rRNA gene fragments in different developmental stages of the same schistosome.
