目的 探讨 p DL12 1的 1.0 kb0 17株问号赖型钩端螺旋体 DNA片段可否作为钩体重组疫苗广谱保护性抗原的候选。方法 采用 SDS- PAGE制备 2 3k Da蛋白 ,免疫日本大耳兔制备抗 2 3k Da抗血清 ,Western blot鉴定其免疫原性。结果 2 3k Da重组抗原兔抗血清可识别 p DL12 1体外表达 2 3k Da抗原产物和问号赖型钩端螺旋体 0 17株超声抗原成份 ;用 Westemblot鉴定抗血清效价 ,其抗体滴度为 1/ 12 80 0 ;用 p DL12 1重组质粒主动免疫豚鼠 ,可使豚鼠抵抗强毒力株钩端螺旋体攻击 ,表现出免疫保护作用。结论 该重组 2 3k Da抗原有良好的免疫原性 ,可能是赖型钩体 0 17株的保护性抗原。
Objective To investigate whether the 1.0 kb0 17 pL121 DNA fragment of p DL12 1 can be used as a candidate for the broad-spectrum protective antigen of recombinant leptospira vaccine. Methods The 23 kDa protein was prepared by SDS-PAGE, and the anti-23 kDa antiserum was prepared by immunizing Japanese big-eared rabbits. The immunogenicity was identified by Western blot. Results 2 3k Da recombinant rabbit anti-rabbit antiserum could recognize 21 kDa antigenic products of p DL12 1 in vitro and 0 17 strains of Leptospira interrogans. The antiserum titer was identified by Westemblot and the titer was 1 / 1280 0; pDL12 1 recombinant plasmid active immunization guinea pigs, guinea pigs can be resistant to virulent strains of leptospiral attack, showing immune protection. Conclusion The recombinant 2 3 k Da antigen has good immunogenicity and may be the protective antigen of 0 17 strain of Leptospira interrogans.
