目的:研究蒺藜提取物对阿霉素(ADR)所致大鼠心肌细胞凋亡的影响。方法:将蒺藜提取物灌胃于ADR所致心肌细胞凋亡的大鼠,采用电镜对大鼠心肌细胞进行超微结构观察,采用TUNEL法检测心肌细胞凋亡指数,免疫组化SP法检测心肌细胞Bax,Bcl-2蛋白的表达。结果:ADR累积用量20 mg·kg-1可致大鼠心肌细胞凋亡,蒺藜提取物中、高剂量组(40,60 mg·kg-1)可减轻心肌细胞超微结构的损伤,蒺藜提取物中、高剂量组较模型组均可显著降低细胞凋亡率(P<0.05)。蒺藜提取物中、高剂量组可显著上调Bcl-2表达(P<0.01)、下调Bax表达(P<0.05)。结论:蒺藜提取物可抑制ADR所致大鼠心肌细胞的凋亡,对阿霉素所致的心脏毒性有保护作用。
Objective: To study the effects of Tribulus terrestris extract on cardiomyocyte apoptosis induced by Adriamycin (ADR) in rats. Methods: The tribulus terrestris extract was intragastrically administered to rat cardiomyocyte apoptosis induced by ADR. The ultrastructure of rat myocardial cells was observed by electron microscopy. The apoptosis index of cardiomyocytes was detected by TUNEL method. Cell Bax, Bcl-2 protein expression. Results: The ADR cumulative dose of 20 mg · kg-1 could induce cardiomyocyte apoptosis in rat myocardial cells. Tribulus terrestris extract high-dose group (40, 60 mg · kg-1) could reduce myocardial ultrastructure damage, Compared with the model group, the apoptosis rates of both medium and high dose groups were significantly decreased (P <0.05). Tribulus terrestris extract, high dose group can significantly up-regulate Bcl-2 expression (P <0.01), down-regulate Bax expression (P <0.05). Conclusion: Tribulus terrestris extract can inhibit rat cardiomyocyte apoptosis induced by ADR and protect cardiotoxicity induced by doxorubicin.
