目的分析缺血性卒中患者血浆葡萄糖脑苷脂酶(GBA)、蛋白磷酸酶2A(PP2A)及其降解产物神经酰胺的改变。方法回顾性纳入2013年5月至9月武警后勤学院附属医院神经内科急性缺血性卒中住院患者45例,另收集同期年龄和性别相匹配的45名体检中心的健康者为对照组。采集患者和健康受试者静脉血,抗凝分离血浆。采用蛋白质芯片H50和激光解析电离飞行时间质谱方法测试血浆神经酰胺水平。结果缺血性卒中患者血浆GBA和PP2A水平均明显低于对照组,缺血性卒中组和对照组的GBA水平分别为(2.4±0.8)、(13.1±1.4)U/L,差异有统计学意义(P<0.05),而PP2A水平分别为(6.5±2.8)、(14.5±4.7)U/L(P<0.01)。缺血性卒中患者血浆神经酰胺相对量为1.9±0.7,明显低于对照组的12.2±5.0(P<0.01)。结论缺血性卒中患者血浆GBA和PP2A以及神经酰胺水平均降低,提示卒中患者血液中α-突触核蛋白的异常磷酸化。
Objective To analyze the changes of plasma glucocerebrosidase (GBA), protein phosphatase 2A (PP2A) and its degradation product ceramide in patients with ischemic stroke. Methods Forty-five inpatients with acute ischemic stroke admitted to Department of Neurology of Affiliated Hospital of Armed Police Forces Hospital were retrospectively collected from May 2013 to September 2013. The other 45 health check-up centers matched with age and gender were collected as control group. Venous blood was collected from patients and healthy subjects, and plasma was separated by anticoagulation. Plasma ceramide levels were tested using protein chip H50 and laser desorption / ionization time-of-flight mass spectrometry. Results The levels of plasma GBA and PP2A in patients with ischemic stroke were significantly lower than those in the control group. The levels of GBA in ischemic stroke group and control group were (2.4 ± 0.8) and (13.1 ± 1.4) U / L, respectively (P <0.05), while the levels of PP2A were (6.5 ± 2.8) and (14.5 ± 4.7) U / L, respectively (P <0.01). The relative amount of plasma ceramide in patients with ischemic stroke was 1.9 ± 0.7, significantly lower than 12.2 ± 5.0 in the control group (P <0.01). Conclusions Plasma levels of GBA, PP2A and ceramide in patients with ischemic stroke are reduced, suggesting abnormal phosphorylation of α-synuclein in the blood of patients with stroke.
