目的 研究PCNA、TGFβ1、CollagenⅢ型在心肌组织的表达及DDPH的作用。方法 用部分狭窄腹主动脉方法 ,从术后第 4周开始 ,ig不同剂量的DDPH (2 5、5 0mg·kg-1·d-1)持续 8wk。用免疫组织化学方法检测PCNA、TGFβ1、CollagenⅢ蛋白表达的变化。 结果 术后 12wk ,发现肥厚组心肌组织PCNA、TGFβ1、CollagenⅢ表达水平分别是对照组的 1 5、1 4和 2 4倍。而DDPH两个给药组蛋白表达水平明显低于肥厚组 ,但仍高于对照组。结论 DDPH对大鼠腹主动脉部分狭窄所致心肌肥厚PCNA、TGFβ1、Col lagenⅢ表达水平的增加有一定逆转作用。
Objective To study the expression of PCNA, TGFβ1 and Collagen Ⅲ in myocardium and the effect of DDPH. Methods With partial narrowing of the abdominal aorta, different doses of DDPH (25, 50 mg · kg-1 · d-1) were administered for 8 weeks starting from the 4th week after operation. The expressions of PCNA, TGFβ1 and Collagen Ⅲ were detected by immunohistochemistry. Results At 12 weeks after operation, the expression of PCNA, TGFβ1 and Collagen Ⅲ in hypertrophic group were found to be 15, 14 and 24 times higher than that in the control group, respectively. The DDPH two administration group protein expression levels were significantly lower than the hypertrophy group, but still higher than the control group. Conclusion DDPH can reverse the increase of the expression of PCNA, TGFβ1 and Col lagen Ⅲ induced by partial stenosis of abdominal aorta in rats.
