目的利用法医DNA标准品对自主研发的法医DNA检验试剂耗材进行电泳检测准确性的实验考查。方法实验平台分为2个:平台1全部由美国Applied Biosystems公司进口的3130xl遗传分析仪、16道电泳毛细管与甲酰胺、电泳缓冲液、凝胶构成;平台2由Applied Biosystems公司进口的3130xl遗传分析仪、16道电泳毛细管与本实验室研发的甲酰胺、电泳缓冲液、凝胶构成。在实验平台上对allelic ladder、内标等标准品以及阳性对照9947a的STR复合扩增产物进行电泳检测,利用GeneMapper软件对电泳结果进行分析。结果自主研发的法医DNA检验试剂耗材可与国外进口的3130xl遗传分析仪配套使用,构建的实验平台对法医DNA标准品的检测结果准确无误。结论自主研发的甲酰胺、电泳缓冲液、凝胶等法医DNA检验试剂耗材检测准确性达到了国外同类产品水平。
Objective To use forensic DNA standards to test the self-developed forensic DNA test reagent consumables for the accuracy of electrophoresis. The experimental platform is divided into two parts: platform 1 is a 3130xl genetic analyzer imported by Applied Biosystems, USA, 16 electrophoresis capillaries are combined with formamide, electrophoresis buffer and gel; Platform 2 is 3130xl imported from Applied Biosystems Instrument, 16 electrophoresis capillary and the laboratory developed formamide, electrophoresis buffer, gel composition. On the experimental platform, the allelic ladder, internal standards and other standards and the positive control 9947a STR complex amplification products were electrophoresis detected using GeneMapper software electrophoresis results were analyzed. Results Independent research and development of forensic DNA testing reagent consumables can be imported with the 3130xl genetic analyzer supporting the use of the experimental platform for the construction of forensic DNA standards accuracy of the test results. Conclusion The self-developed forensic DNA testing reagents such as formamide, electrophoresis buffer and gel have reached the same foreign product accuracy.
