目的总结成纤维激活蛋白(fibroblast activation protein,FAP)的功能、作用机制以及其在靶向治疗方面的应用。方法复习近年来国内外有关FAP的相关研究文献并进行综述。结果 FAP与人二肽基肽酶Ⅳ具有重要的联系,具有丝氨酸蛋白酶活性,是肿瘤微环境中重要的免疫抑制成分。FAP参与肿瘤性和非肿瘤性疾病的病理过程。在靶向治疗中,针对FAP的酶活性抑制物、抗体、疫苗及前体药物被广泛研究。结论 FAP功能多样,参与多种疾病的病理过程,同时对肿瘤靶向治疗的研究具有重要意义。
Objective To summarize the function and mechanism of fibroblast activation protein (FAP) and its application in targeted therapy. Methods To review and summarize the related literatures about FAP at home and abroad in recent years. Results FAP has an important relationship with human dipeptidyl peptidase Ⅳ and has serine protease activity. It is an important immunosuppressive component in the tumor microenvironment. FAP is involved in the pathological processes of both neoplastic and non-neoplastic diseases. Inhibitors of the enzyme activity against FAP, antibodies, vaccines and prodrugs are widely studied in targeted therapy. Conclusion FAP is diverse in functions and involved in the pathological process of many diseases. It is also of great significance for the study of tumor targeted therapy.
