目的研究白花蛇舌草对CEM细胞抑制作用及机制。方法以不同浓度的白花蛇舌草水提物作用CEM细胞24,48,72h后,通过MTT比色试验,检测其对CEM细胞的抑制率;用浓度分别为166μg/ml、33.2μg/ml、6.64μg/ml的白花蛇舌草水提物作用于CEM细胞24,48,72h后,通过逆转录PCR(RT-PCR)检测P53基因的表达,探索白花蛇舌草抑制CEM细胞的可能机制。结果白花蛇舌草水提物能够抑制CEM细胞的生长;白花蛇舌草能促进CEM细胞的P53基因表达,且随用药浓度的升高及作用时间的延长,其抑制作用逐渐增强、P53基因表达也逐渐增强。结论白花蛇舌草水提物能抑制白血病CEM细胞的生长,其作用强度与时间、浓度呈正相关,其抑制机制可能与上调P53基因的表达有较大关联。
Objective To study the inhibitory effect and mechanism of Hedyotis diffusa Willd on CEM cells. Methods CEM cells were treated with aqueous extracts of Hedyotic diffusa at different concentrations for 24, 48, and 72 hours. MTT colorimetric assay was used to determine the inhibitory rate of CEM cells. The concentrations of CMA were 166μg / ml, 33.2μg / ml, After the HHD of 6.64μg / ml was treated for 24, 48 and 72h in CEM cells, the expression of P53 gene was detected by reverse transcription-polymerase chain reaction (RT-PCR) to explore the possible mechanism of inhibition on CEM cells. Results Hedyotis diffusa aqueous extract could inhibit the growth of CEM cells. Hedyotis diffusa could promote the expression of P53 gene in CEM cells. With the increase of drug concentration and prolongation of action time, the inhibitory effect of Hedyotis diffusa Willd on the expression of P53 gene Also gradually increased. Conclusion The aqueous extract of Hedyotis diffusa can inhibit the growth of leukemia CEM cells. The intensity of action is positively correlated with time and concentration, and its inhibitory mechanism may be related to the up-regulation of the expression of P53 gene.
