目的研究体外建立模拟CO2、He气腹环境对胃癌MKN-45细胞增殖的影响,初步探讨CO2气腹腹腔镜胃癌手术的安全性。方法体外建立模拟CO2、He气腹环境,全自动气腹机维持压力为12mmHg,作用3h并没对照组。处理后0、0.5、1、1.5、2、2.5、3h,用pH计检测细胞培养液pH值变化;用MTT法检测第1、2、3、4、5、6、7天细胞增殖情况。结果处理结束后CO2气腹组细胞培养液pH值呈弱酸性,与对照组比下降非常显著(P<0.01),处理后2hpH值恢复正常;He气腹组pH值呈弱碱性,与对照组比升高显著(P<0.05),1.5h恢复正常。MTT法检测细胞增殖结果显示,CO2气腹组和He气腹组人胃癌MKN-45细胞的增殖与对照组比无统计学差异(P>0.05)。结论体外模拟12mmHgCO2气腹和He气腹持续作用3h时,均不促进胃癌细胞增殖。
Objective To study the effects of CO2 and He pneumoperitoneum environment on the proliferation of gastric cancer MKN-45 cells in vitro and to explore the safety of CO2 pneumoperitoneum laparoscopic gastric cancer surgery. Methods The CO2 pneumonia and pneumoperitoneum were established in vitro. The pressure of automatic pneumoperitoneum was maintained at 12mmHg. No effect was observed after 3 hours of treatment. After treated for 0, 0.5, 1, 1.5, 2, 2.5 and 3h, the pH value of cell culture medium was detected by pH meter. The proliferation of cells was detected by MTT assay on day 1,2,3,4,5,6,7. Results After the treatment, the pH value of CO2 pneumoperitoneum cell culture solution was weakly acidic, which was significantly lower than that of the control group (P <0.01), and returned to normal after 2 hours of treatment. The pH value of He pneumoperitoneum group was weakly alkaline, Group increased significantly (P <0.05), 1.5h returned to normal. The results of MTT assay showed that there was no significant difference in the proliferation of MKN-45 cells between the CO2 pneumoperitoneum group and the He pneumoperitoneum group and the control group (P> 0.05). Conclusion In vitro simulation of 12mmHgCO2 pneumoperitoneum and He pneumoperitoneum continued for 3h, they did not promote the proliferation of gastric cancer cells.
