纤溶酶原激活物抑制剂1(PAI-1)是一种重要促凝血因子。内毒素脂多糖(LPS)通过刺激血管内皮细胞(EC)分泌PAI-1且而导致血栓形成。采用ELISA的Northern印迹方法观察中药川芎嗪对EC及LPS致EC表达PAI-1的影响。结果表明川芎嗪不仅抑制LPS致EC PAI-1的分泌和mRNA表达,而且抑制 EC PAI-1基础水平mRNA表达。川芎嗪对LPS致 EC PAI-1分泌的抑制呈剂量效应关系。为川芎嗪抗感染性休克和治疗心血管疾病提供了重要的理论和实验依据。
Plasminogen activator inhibitor 1 (PAI-1) is an important procoagulant. Lipopolysaccharide (LPS), an endotoxin, secretes PAI-1 by stimulating vascular endothelial cells (ECs) and leads to thrombosis. The effect of Ligustrazine on EC expression of PAI-1 induced by EC and LPS was observed by Northern blotting with ELISA. The results showed that ligustrazine not only inhibited LPS-induced EC PAI-1 secretion and mRNA expression, but also inhibited EC PAI-1 basal level mRNA expression. Ligustrazine inhibited LPS-induced EC PAI-1 secretion in a dose-response relationship. It provided important theoretical and experimental evidences for tetramethylpyrazine against septic shock and treatment of cardiovascular diseases.
