采用差速离心和20~45%Renografin梯度离心方法从Rickettsic感染卵黄囊膜中获得了较高纯度的Rickettsic颗粒。蛋白回收0.716mg/个膜,粒子数回收66.5%。有两种密度的颗粒分别为1.220和1.244。0.1%福尔马林灭活条件较56℃灭活对Rickettsia的提纯影响小。灭活后Rickettsic两种颗粒浮力密度稍有改变,分别为1.210和1.225。
Rickettsic particles with higher purity were obtained from Rickettsic-infected yolk sac membranes by differential centrifugation and gradient 20-45% Renografin centrifugation. Protein recovery 0.716mg / membrane, the number of particles recovered 66.5%. The two-density particles were 1.220 and 1.244.0.1% respectively. The inactivation of formalin had less effect on the purification of Rickettsia than the 56 ℃ inactivation. The buoyant density of Rickettsic particles changed slightly after inactivation, at 1.210 and 1.225, respectively.
