采用ISSR标记技术对40个姬菇杂交菌株及2个亲本菌株进行分析,使用NTSYSpc2.1生物软件对42个供试菌株进行聚类分析,构建系统树。试验筛选出了11个扩增谱带清晰、多态性好的ISSR引物,共扩增出81条清晰易辨的多态性谱带。聚类分析结果表明,42个供试菌株遗传相似系数变异范围为0.4286~0.8537,在相似系数0.6680时,42个菌株聚为7类,两亲本各聚一类,杂交菌株与亲本菌株遗传差异较大。
ISSR markers were used to analyze 40 hybrid strains of hyoscyamus and two parental strains. Cluster analysis was performed on 42 tested strains using NTSYSpc2.1 biological software to construct a phylogenetic tree. Eleven ISSR primers with clear and polymorphic amplification bands were screened out and 81 clear and legible bands were amplified. The results of cluster analysis showed that the genetic similarity coefficients of 42 tested strains ranged from 0.4286 to 0.8537. When the similarity coefficient was 0.6680, 42 strains clustered into 7 categories, and the two parents clustered in one class. The genetic differences between the hybrid strains and the parental strains were relatively Big.
