目的利用血清药理学的方法,观察荔枝核含药血清体外的抑制肿瘤细胞生长;并研究荔枝核对小鼠S180,EAC体内、外细胞生长及凋亡作用。方法小鼠S180,EAC细胞混悬液用生理盐水按1:1进行稀释制成含瘤腹水混悬液,在给药前24 h,每只小鼠腋窝皮下接种0.2 ml。荔枝核水提液对小鼠S180,EAC抑瘤实验连续给药10 d。测定肿瘤作用与细胞凋亡表达。结果荔枝核提取物30%含药血清高中剂量和20%含药血清高剂量对HepG2肿瘤细胞具有明显的抑瘤作用,Hochest染色肿瘤细胞出现凋亡形态;而流式细胞仪检测可见30%含药血清高中低剂量的促肿瘤凋亡率分别是63.3%,45.5%和23.8%,明显高于正常血清组。荔枝核水提物能够明显抑制S180,EAC肿瘤的生长(P<0.05),荔枝核提取物对荷S180小鼠高剂量组的抑瘤率达到32.81%。对荷EAC实体瘤小鼠的肿瘤其抑瘤率达到30.62%;结论荔枝核具有抑制小鼠S180,EAC体内、外细胞生长的影响,促进肿瘤细胞的凋亡,从而表现抗肿瘤的作用。
OBJECTIVE: To use serum pharmacology methods to observe the inhibitory effect of litchi nucleus-containing serum on the growth of tumor cells in vitro and to investigate the effects of litchi nucleus on the growth and apoptosis of S180 and EAC cells in vivo and in vitro. Methods Mouse S180 and EAC cell suspensions were diluted with 1:1 saline solution to make a tumor-bearing ascites suspension. Each mouse was inoculated subcutaneously with 0.2 ml 24 h before administration. Litchi seed aqueous extracts were continuously administered to mice for S180, EAC tumor inhibition experiments for 10 days. The tumor effect and apoptosis expression were measured. Results The high dose of 30% drug-containing serum and the high dose of 20% drug-containing serum in Litchi seed extract had significant anti-tumor effect on HepG2 tumor cells. Hochest stained tumor cells with apoptotic morphology; 30% were detected by flow cytometry. The tumor-promoting apoptotic rates of high, medium, and low doses of drug serum were 63.3%, 45.5%, and 23.8%, respectively, which were significantly higher than those of the normal serum group. Litchi seed aqueous extract significantly inhibited the growth of S180 and EAC tumors (P<0.05). The inhibitory rate of Litchi seed extract against high dose S180 mice was 32.81%. The tumor-inhibitory rate of tumors of mice bearing EAC solid tumors reached 30.62%. Conclusion Litchi nucleus inhibits the growth of S180 and EAC cells in vivo and in vitro, and promotes the apoptosis of tumor cells, thereby demonstrating the anti-tumor effect.
