本文采用吞噬发光技术(CL)测定RA诱导及未诱导的HL-60细胞的吞噬氧化能力,并与NBT还原反应进行比较,以此作为判定HL-60细胞诱导成熟的一个功能性指标。总结其规律性为:(1)未诱导的HL-60细胞无CL,而RA可以诱导HL-60分化成熟,产生CL。在一定浓度范围内(10~(-6)~10~(-6)M),CL值随着RA浓度增加而增高;(2)在我们所使用不同浓度的RA诱导无换浓培养的HL-60细胞时,其CL值增减规律相同。开始随着诱导时间延长,CL值增加,在第4天达到最大值。细胞生长曲线的峰值也是在第3、4天。还原NBT的细胞的百分比也基本循此规律。不同之处为,第5天CL值下降,而NBT%仍然增高;(3)CL曲线峰值出现的速率随着诱导天数的增加而逐渐加快,即达到峰值的时间缩短。
In this study, phagocytosis (CL) was used to determine the phagocytic activity of RA-induced and non-induced HL-60 cells and compared with the NBT reduction reaction as a functional index for the induction of maturation of HL-60 cells. To summarize its regularity: (1) non-induced HL-60 cells without CL, and RA can induce differentiation and maturation of HL-60 to produce CL. In a certain concentration range (10 -6 -6 10 M), CL value increased with the increase of RA concentration; (2) We used different concentrations of RA induced HL without exchange culture HL -60 cells, the CL value of the same increase and decrease. Starting with the induction time, the CL value increased, reaching its maximum on the fourth day. The peak of cell growth curve is also in the first 3,4 days. The percentage of cells that restore NBT also follows this pattern. The difference is that on the fifth day, the CL value decreased and the NBT% increased. (3) The rate of peak value of CL curve gradually accelerated with the increase of induction days, that is, the peak time was shortened.
