[目的]通过检测Vav1、Ras相关的C3肉毒素底物1(Rac1)、细胞周期蛋白D1(Cyclin D1)、p21蛋白在结直肠癌组织中的表达情况,分析Vav1蛋白与Rac1、Cyclin D1、p21的关系及意义。[方法]180例结直肠癌肿瘤组织和162例癌旁组织的石蜡组织标本进行免疫组化染色,检测肿瘤组织和癌旁组织中Vav1、Rac1、Cyclin D1、p21蛋白表达。[结果]结直肠癌肿瘤组织中Vav1、Rac1、Cyclin D1蛋白表达高于癌旁组织,p21蛋白在癌组织表达低于癌旁组织(P<0.05)。Vav1蛋白表达与肿瘤的浸润深度、分化情况及淋巴结转移有关;Rac1与肿瘤的分化情况、淋巴结转移有关;Cyclin D1表达与肿瘤的浸润深度、淋巴结转移有关(均P<0.05)。Spearman相关分析显示,肿瘤组织中Vav1蛋白表达与Rac1、Cyclin D1蛋白呈正相关,与p21负相关(P<0.05)。[结论]Vav1在结直肠癌组织中的表达与肿瘤增殖的指标有关,可能通过调节Rac1、Cyclin D1、p21促进肿瘤的生长。
[Objective] The purpose of this study was to investigate the expression of Vav1, Rac1, Cyclin D1 and p21 in colorectal cancer tissues. The expressions of Vav1, Rac1, Cyclin D1, p21 the relationship and significance. [Method] Paraffin tissue specimens from 180 cases of colorectal cancer tissue and 162 cases of para-cancerous tissue were immunohistochemically stained to detect the expression of Vav1, Rac1, Cyclin D1 and p21 in tumor tissues and adjacent tissues. [Results] The protein expressions of Vav1, Rac1 and Cyclin D1 in colorectal cancer tissues were higher than those in paracancerous tissues, while p21 protein was lower in cancer tissues than in paracancerous tissues (P <0.05). The expression of Vav1 was related to the depth of tumor invasion, differentiation and lymph node metastasis. Rac1 was related to tumor differentiation and lymph node metastasis. The expression of Cyclin D1 was related to the depth of invasion and lymph node metastasis (all P <0.05). Spearman correlation analysis showed that the expression of Vav1 protein in tumor tissues was positively correlated with Rac1 and Cyclin D1 proteins, and negatively correlated with p21 (P <0.05). [Conclusion] The expression of Vav1 in colorectal cancer tissues is related to tumor proliferation, which may promote tumor growth by regulating Rac1, Cyclin D1 and p21.
