目的探讨强心甙类药物洋地黄毒甙或地高辛对长春新碱抗肿瘤活性的协同作用。方法台盼蓝排染法测定药物对K562细胞的杀伤作用;用小鼠肝癌腹水瘤H22模型,观察药物对小鼠生命延长率的影响。结果洋地黄毒甙、地高辛能显著杀伤K562细胞,并有剂量依赖性,IC50分别为0.059,0.101μg/ml;低浓度洋地黄毒甙或地高辛对低浓度长春新碱杀伤K562细胞呈现增强作用(q>1)。洋地黄毒甙或地高辛对长春新碱抑制H22腹水瘤也呈增强作用(g>1)。结论洋地黄毒甙、地高辛对长春新碱抗肿瘤活性有增强作用。
Objective To investigate the synergistic effect of cardiac glycosides digoxin or digoxin on the antitumor activity of vincristine. Methods Trypan blue exclusion assay was used to determine the killing effect of K562 cells on K562 cells. The H22 model of mouse ascites tumor was used to observe the effects of drugs on the mouse survival rate. Results Digoxin and digoxin could significantly kill K562 cells in a dose-dependent manner with IC50 of 0.059 and 0.101 μg / ml, respectively. Low concentration of digoxin or digoxin could inhibit the proliferation of K562 cells at low concentration of vincristine Killing K562 cells showed enhanced (q> 1).
