目的探讨氢醌(HQ)对TK6细胞白血病相关miRNA表达的影响。方法以磷酸盐缓冲液(PBS)溶解HQ,以PBS组为对照组,分别以2.5、5.0、10.0和20μmol/L HQ染毒TK6细胞48 h。应用反转录实时荧光定量-聚合酶链反应检测miR-126、miR-155、miR-196b和miR-20b的表达改变。结果与对照组相比,HQ对TK6细胞的上述4种白血病相关miRNA的表达改变均有影响,miR-126的表达上调,miR-155、miR-196b和miR-20b的表达均为下调,且均以20μmol/L HQ组最为明显,miR-126、miR-155、miR-196b和miR-20b分别上调83%,下调40%,49%和42%(P<0.05)。结论 HQ能促进miR-126的表达,抑制miR-155、miR-196b和miR-20b的表达。
Objective To investigate the effect of hydroquinone (HQ) on miRNA expression in leukemia of TK6 cells. Methods HQ was dissolved in phosphate buffered saline (PBS) and the control group was treated with PBS. The TK6 cells were exposed to 2.5, 5.0, 10.0 and 20 μmol / L HQ for 48 h. The expression of miR-126, miR-155, miR-196b and miR-20b were detected by reverse transcription real-time quantitative polymerase chain reaction. Results Compared with the control group, HQ had an effect on the expression changes of the four leukemia-associated miRNAs in TK6 cells, the expression of miR-126 was up-regulated, the expression of miR-155, miR-196b and miR- MiR-126, miR-155, miR-196b and miR-20b were up-regulated by 83%, 40%, 49% and 42% respectively (P <0.05) in 20μmol / L HQ group. Conclusion HQ can promote the expression of miR-126 and inhibit the expression of miR-155, miR-196b and miR-20b.
